Step 1 – Solution preparation

In two 1.5 mL microcentrifuge tubes (labeled GUS and CK):

Add 1 mL GUS staining buffer

Add 20 μL X-gluc solution (colorless = effective; red = degraded)

Mix gently

Step 2 – Seed treatment

Remove husks from rice seeds

Cut each seed in half along the short axis with a blade

Step 3 – Incubation

Incubate overnight at 25°C (room temperature)

Step 4 – Expected results

Seed 01: Blue coloration.

Seed 02: No blue coloration.
